Endogenous Human TLR3 Functional Cellular Assay

Target: Toll-Like Receptor 3 (TLR3)

Cell Line: A549 (human lung carcinoma cells). A549 cells have been shown to express TLR3 but appear to be devoid of TLR7 and TLR8.[1]

TLRs aremembers of a larger Interleukin-1 receptor superfamily and play a key role in the innate immune response and pathogen recognition.[2] TLRs are expressed on a variety of immune cells, such as neutrophils,macrophages and dendritic cells and work by recognizing specific conserved motifs on invading pathogens.With 10 members discovered to-date, the TLR family represent an attractive class of therapeutic targets and have been implicated in several pathophysiological conditions including cancer, inflammation, autoimmune and infectious diseases.[3]


Dose-dependent activation of endogenous TLR3 in A549 cells by Poly (I-C) (Panel A). The PI3K inhibitor LY29004 potentiates the Poly (I-C)mediated activation of TLR3 indicating a repressive role of PI3K on human TLR3 (Panel B).
