Absorbance Detection Technologies
Authored by: Paul Taylor, Boehringer Ingleheim Inc.
In colorimetric assays, a reagent is added to a solution of the substance that is to be quantitatively analyzed. The color that develops is "read" by measuring the absorbance with a photometer. The properties of absorbance-based assays are described by the Beer-Lambert Law and care must be taken to avoid compound quenching and/or saturation effects which drive signal responses into a non-linear range.
